In this study, total phenolic and flavonoid contents, antioxidant activity and cytotoxic effects of hexane
and methanol extracts obtained from the aerial parts of Hypericum linarioides Bosse (H. linarioides)
were investigated. Total phenolic and total flavonoid contents of the extracts were determined by Folin
ciocalteu and aluminum chloride colorimetric analysis, respectively. Antioxidant activity was evaluated
by DPPH radical scavenging and iron ion reducing antioxidant power (FRAP) assays. The cytotoxicity
of the extracts was investigated by XTT assay on human breast adenocarcinoma (MCF-7) cell line. Folin
ciocalteu and aluminum chloride studies showed that methanolic extract was richer in phenolic and
flavonoid contents. In parallel to phenolic contents, methanolic extract exhibited much higher DPPH
radical scavenging and iron ion reduction capacity. XTT analysis results revealed that the methanolic
extract exhibited much stronger cytotoxicity against the tested MCF-7 cancerous cells with its much
lower IC50 values. In conclusion, all these results suggest that H. linarioides may be an important basis
for using as an alternative to chemotherapeutic drugs used in breast cancer treatment with less side
effects and also can be a source for other preclinical studies to be conducted with this species.
