Özet

Background: Doxorubicin (DOX) is an antineoplastic agent known for its testicular

toxicity side effect. Mechanisms of DOX-induced testicular damage include inflammation, reactive

oxygen and nitrogen species, and oxidative stress. Centella asiatica (CA) has anti-inflammatory

and antioxidant effects. The purpose of the study is to investigate the protective effects of Centella

asiatica extract aganist DOX-induced testicular toxicity in rats. Methods: The rats were randomly

assigned as four group; Healthy control (HG), Centella asiatica extract (CA), doxorubicin (DOX),

CA extract + DOX (CADX). CA extract 200 mg/kg was given orally the CA and CADX groups for

seven days. The HG and DOX groups received oral saline. One hour after, DOX 7.5 mg/kg

administered (intraperitoneally) DOX and CADX groups on days 1, 4, and 7. After that, rats were

euthanized with thiopental sodium, and tissues were removed. Oxidant/antioxidant, nitric oxide,

and cytokine levels in the tissue and hormone levels in the blood were investigated. Results: CA

extract significantly protected the DOX-induced elevation of malondialdehyde, nitric oxide, and

pro-inflammatory cytokines and reduction of superoxide dismutase and hormones. CA reduced

the severity of DOX destruction, oedema, dilatation/congestion blood vessels, connective tissue,

hemorrhage, and PMNL infiltration damage in testicular tissue from grade 3 to grade 1. Moreover,

it improved reproductive hormone dysfunction. Conclusion: CA may be beneficial in treating DOXrelated

male reproductive organ injury and dysfunction.
